Hepatitis C Virus Down-Regulates the Expression of Ribonucleotide Reductases to Promote Its Replication

چکیده
Ribonucleotide reductases (RRs or RNRs) catalyze the reduction of OH group on 2nd carbon ribose, reducing four ribonucleotides (NTPs) to corresponding deoxyribonucleotides (dNTPs) promote DNA synthesis. Large viruses, such as herpesviruses and poxviruses, could benefit their replication through increasing dNTPs via expression viral RRs. Little is known regarding relationship between cellular RRs RNA viruses. Mammalian contain two subunits ribonucleotide reductase M1 polypeptide (RRM1) M2 (RRM2). In this study, RRMs, including RRM1 RRM2, found be down-regulated in hepatitis C virus (HCV)-infected Huh7.5 cells Huh7 with HCV subgenomic RNAs (HCVr). As expected, NTP/dNTP ratio elevated HCVr cells. Compared that control sh-scramble, RRM-knockdown elevated. Knockdown RRM2 increases replicon Moreover, inhibitors Didox, Trimidox hydroxyurea, enhance replication. Among various proteins, NS5A and/or NS3/4A proteins suppress RRMs. When these are taken together, results suggest down-regulates RRMs cultured its

Virus-Encoded Ribonucleotide Reductases
Ribonucleotide reductases [EC 1.17.4.] are a group of enzymes which critically contribute to the biosynthesis of deoxyribonucleotides (dNTPs), the monomeric precursors of DNA polymerization. By a radical reaction mechanism, they catalyze the reduction of the ribonucleotides, ADP, GDP, CDP, and UDP, mostly at the diphosphate level, to their deoxy conterparts dADP, dGDP, dCDP, and dUDP, respectiv...
متن کاملPyruvate dehydrogenase kinase regulates hepatitis C virus replication
During replication, hepatitis C virus (HCV) utilizes macromolecules produced by its host cell. This process requires host cellular metabolic reprogramming to favor elevated levels of aerobic glycolysis. Therefore, we evaluated whether pyruvate dehydrogenase kinase (PDK), a mitochondrial enzyme that promotes aerobic glycolysis, can regulate HCV replication. Levels of c-Myc, hypoxia-inducible fac...

متن کاملHepatitis C Virus Increases Free Fatty Acids Absorption and Promotes its Replication Via Down-Regulating GADD45α Expression
BACKGROUND Hepatitis C virus (HCV) infection, as a major cause of chronic hepatic diseases, is always accompanied with an abnormality of lipid metabolism. The aim of this study was to investigate the pathogenic role of free fatty acids (FFA) in human HCV infection. MATERIAL AND METHODS Peripheral blood lipid indexes among HCV patients with different viral loads (199 samples) and healthy donors ...
متن کاملActive RNA Replication of Hepatitis C Virus Downregulates CD81 Expression
So far how hepatitis C virus (HCV) replication modulates subsequent virus growth and propagation still remains largely unknown. Here we determine the impact of HCV replication status on the consequential virus growth by comparing normal and high levels of HCV RNA expression. We first engineered a full-length, HCV genotype 2a JFH1 genome containing a blasticidin-resistant cassette inserted at am...
